    Andrographolide: A Novel Antimalarial Diterpene Lactone Compound from Andrographis paniculata and Its Interaction with Curcumin and Artesunate

    Andrographolide (AND), the diterpene lactone compound, was purified by HPLC from the methanolic fraction of the plant Andrographis paniculata. The compound was found to have potent antiplasmodial activity when tested in isolation and in combination with curcumin and artesunate against the erythrocytic stages of Plasmodium falciparum in vitro and Plasmodium berghei ANKA in vivo. IC50s for artesunate (AS), andrographolide (AND), and curcumin (CUR) were found to be 0.05, 9.1 and 17.4 ΌM, respectively. The compound (AND) was found synergistic with curcumin (CUR) and addictively interactive with artesunate (AS). In vivo, andrographolide-curcumin exhibited better antimalarial activity, not only by reducing parasitemia (29%), compared to the control (81%), but also by extending the life span by 2-3 folds. Being nontoxic to the in vivo system this agent can be used as template molecule for designing new derivatives with improved antimalarial properties

    Pool fires are the common consequences of accidentally released flammable liquid fuels. Such accidents can occur in a storage facility or during the transportation of hazardous substances. The safety distances are therefore important to be measured for the safe storage and transportation of such flammable liquids. Organic peroxides are energetic materials, capable to decompose and are highly reactive in nature. Therefore, the safety from their hazard is a great concern for the chemical industries and regulating authorities. In the present PhD work the safety distances from two organic peroxides namely tert-butyl peroxybenzoate (TBPB) and tert-butyl ethylhexanoate (TBPEH) pool fires are experimentally investigated and predicted with CFD simulation.Pool BrĂ€nde sind die hĂ€ufigsten Folgen von versehentlich freigegeben brennbare flĂŒssige Kraftstoffe. Solche UnfĂ€lle können in einer LagerstĂ€tte vorkommen oder wĂ€hrend des Transports von gefĂ€hrlichen Stoffen. Die SicherheitsabstĂ€nde sind daher wichtig fĂŒr die sichere Lagerung und Transport von brennbaren FlĂŒssigkeiten wie gemessen werden. Organische Peroxide sind energetische Materialien, zu zerlegen und in der Lage sind hochreaktive in der Natur.     Boiling Liquid Expanding Vapour Explosion (BLEVE) of Peroxy-fuels: Experiments and Computational Fluid Dynamics (CFD) Simulation

    AbstractFire and explosion hazards associated with storage and transportation of flammable materials have been a matter of great interest in the recent times. BLEVE is a scenario that occurs when a closed fuel container is subjected to heat for a longer duration. Such events are disastrous to human beings and assets both. In the past there have been numerous studies on BLEVEs and fireballs of hydrocarbon fuels, e.g. kerosene, gasoline, LPG, LNG and others. Though, the fireballs of peroxy-fuels are not looked into detail as such. This article tries to overcome this lack of knowledge. Both, experimental investigation and CFD simulations are performed to measure and predict the fireball characteristics of a peroxy-fuel. Due to thermal decomposition in the liquid phase and active oxygen content a peroxy-fuel fireball burns at a very fast rate and emit higher thermal radiation whereas exhibits smaller diameter and elevation compared to hydrocarbons. That eventually leads to consideration of larger safety distances from them which are also verified by CFD results

    Anti-malarial activities of Andrographis paniculata and Hedyotis corymbosa extracts and their combination with curcumin

    <p>Abstract</p> <p>Background</p> <p>Herbal extracts of <it>Andrographis paniculata </it>(AP) and <it>Hedyotis corymbosa </it>(HC) are known as hepato-protective and fever-reducing drugs since ancient time and they have been used regularly by the people in the south Asian sub-continent. Methanolic extracts of these two plants were tested in vitro on choloroquine sensitive (MRC-pf-20) and resistant (MRC-pf-303) strains of <it>Plasmodium falciparum </it>for their anti-malarial activity.</p> <p>Methods</p> <p>Growth inhibition was determined using different concentrations of these plant extracts on synchronized <it>P. falciparum </it>cultures at the ring stage. The interactions between these two plant extracts and individually with curcumin were studied in vitro. The performance of these two herbal extracts in isolation and combination were further evaluated in vivo on Balb/c mice infected with <it>Plasmodium berghei </it>ANKA and their efficacy was compared with that of curcumin. The in vivo toxicity of the plant derived compounds as well as their parasite stage-specificity was studied.</p> <p>Results</p> <p>The 50% inhibitory concentration (IC<sub>50</sub>) of AP (7.2 ÎŒg/ml) was found better than HC (10.8 ÎŒg/ml). Combination of these two herbal drugs showed substantial enhancement in their anti-malarial activity. Combinatorial effect of each of these with curcumin also revealed anti-malarial effect. Additive interaction between the plant extracts (AP + HC) and their individual synergism with curcumin (AP+CUR, HC+CUR) were evident from this study. Increased in vivo potency was also observed with the combination of plant extracts over the individual extracts and curcumin. Both the plant extracts were found to inhibit the ring stage of the parasite and did not show any in vivo toxicity, whether used in isolation or in combination.</p> <p>Conclusion</p> <p>Both these two plant extracts in combination with curcumin could be an effective, alternative source of herbal anti-malarial drugs.</p

    One of the most striking characteristic of the brain is its profuse neuronal connectivity. Not surprisingly, the function of the nervous system critically depends on the spatiotemporal pattern of intercommunication between different regions of the brain. Both macro- and microscopic aspects of the wiring diagrams of brain circuits are relevant and need to be understood in order to cope with the complexity of the brain function. In this way, for instance, the long-range connections that carry the functional specification of cortical territories need to be studied together with the detailed microcircuits inside a cortical column. Moreover, the temporal dimension of these wiring diagrams must be investigated since neuronal networks are dynamic structures exhibiting context-dependent changes in synaptic weights (Canals et al., 2009) and numbers (Chklovskii et al., 2004). Investigations over the last decades strongly suggest that stimulus or task related neural activity is distributed over large parts of the brain, covering different cortical and sub-cortical areas. For a detailed understanding of brain function, it is of prime importance to understand the organization of the neuronal connections. To chart the anatomical connections between the various components of brain networks, the neuronal tract tracing technique has been proved to be very useful. Thus, experimental tools that allow the exploration of brain circuits at diverse organizational levels are mandatory for the understanding of brain intercommunication and information processing

    Sentiment analysis and classification of Indian farmers’ protest using twitter data

    Protests are an integral part of democracy and an important source for citizens to convey their demands and/or dissatisfaction to the government. As citizens become more aware of their rights, there has been an increasing number of protests all over the world for various reasons. With the advancement of technology, there has also been an exponential rise in the use of social media to exchange information and ideas. In this research, we gathered data from the microblogging website Twitter concerning farmers’ protest to understand the sentiments that the public shared on an international level. We used models to categorize and analyze the sentiments based on a collection of around 20,000 tweets on the protest. We conducted our analysis using Bag of Words and TF-IDF and discovered that Bag of Words performed better than TF-IDF. In addition, we also used Naive Bayes, Decision Trees, Random Forests, and Support Vector Machines and also discovered that Random Forest had the highest classification accuracy
